ABSTRACT

We investigated the effects of the juçara fruit (Euterpe edulis Martius) pulp and lyophilized extract on the expression of cytoprotective genes nuclear factor erythroid 2 (NF-E2)-related factor 2 (NRF2), kelch-like ECH-associated protein 1 (KEAP1), superoxide dismutase (SOD1), and glutathione peroxidase (GPX2) in human colorectal cancer cell lines (HT-29 and Caco-2). Cells were cultured for 24 h in Dulbecco's Modified Eagle's Medium containing juçara fruit pulp (5, 10, or 50 mg/mL) or lyophilized extract (0.05, 0.1, or 0.5 mg/mL), and gene expression was quantified using real-time quantitative reverse transcription polymerase chain reaction. All studied genes showed significant variation in gene expression among different concentrations of pulp or lyophilized extract. Overall, the expression of the selected genes decreased in both cell lines following exposure to the pulp or lyophilized extract in a dose-dependent manner for most of the concentrations studied. In summary, our study showed that the compounds in juçara fruit inhibited the expression of cytoprotective genes associated with the antioxidant response and that, although not cytotoxic at the concentrations studied, they could potentially block the activation of the NRF2/KEAP1 pathway.

ABSTRACT

Troponin I as a biomarker of cardiac injury in sepsis, and other hematological parameters in female dogs with pyometra were evaluated. The groups did not differ in the assessment of serum cardiac troponin I. The total amount of leukocytes (mm3) and percentage of band cells was significantly higher in the sepsis group (23,221.74±16,848.80mm3 and 5.91±10.18%) compared to the non-sepsis group (14,492.86±6,828.26mm3 and 1.93±1.64%) and the control group (10,320.00±3,999.02mm3 and 1.65±2.05% respectively). There were significant differences in serum concentrations of C-reactive protein (mg / dL) in the sepsis group (19.57±41.69md / dL) compared to the non-sepsis group (10.29±12.02mg / dL) and control group (3.60±3.53mg / dL). In the evaluation of serum lactate concentration, there was a significant difference between dogs with pyometra and healthy dogs, but there was no significant difference between the sepsis and non-sepsis groups. The results of this study indicate that troponin I could not be considered an early biomarker of myocardial injury in cases of dogs with pyometra, because the results of the measurements were similar between groups, inferring that there may not have been an cardiomyocytes injury at this stage. C-reactive protein and lactate are potential markers for systemic inflammation, as demonstrated by significantly higher serum concentrations in dogs with pyometra.(AU)

ABSTRACT

The aim of this trial was to study the effect of Growth Hormone (GH) on the seminal parameters and testosterone profile from Nellore bulls. Sixteen bulls were allocated in a 2 x 2 factorial arrangement (two ages and two r-bST levels), with four animals per treatment. The r-bST was injected every 14 days, with a total of nine injections per animal, during the 120 days of the experimental period. The semen collections were done every 15 days and blood samples were collected in four hour intervals for 24 hours, and in 30 day intervals from the first r-bST injection for testosterone profile. The semen of adult bulls treated with r-bST showed greater MOT and VIG (P<0.05) in relation to the control (79.70±11.56% vs 64.06±23.65%) and (3.80±0.88 vs 2.92±1.25) respectively; however, the other evaluated seminal parameters were not affected (P>0.05) by the r-bST treatment. The physical measurements of the semen of young animals was also unaffected (P>0.05) by r-bST injection. However, the treatment with r-bST decreased the number of minor morphologic defects (narrow head, giant head, small normal head, round head, free normal head, abaxial implantation, simple bent tail and distal droplet) compared to untreated young bulls (P<0.05). The serum testosterone concentration was influenced (P<0.05) by treatment, at 120 days after the first injection of r-bST and they showed a tendency of three picks in a 24 hour period of sampling.

ABSTRACT

The productive and economic performance of different beef genetic groups in a pasture-based growing system and feedlot finishing with one year cycle was evaluated. Thirty-six bulls from three groups F1Guzerat x Nellore F1 (GN); F1 Guzerá x Hosltein (GH) and F1 1/2 Simmental + 1/4 Guzerá + 1/4 Nellore (SG), with average age of 270 (± 30) days and average body weight of 241.3 (± 34.0) kg were utilized. The experimental design utilized was completely randomized (3 x 12). During the growing phase, bulls were kept in rotational Brachiaria brizantha cv Marandu pastures, receiving commercial supplement; in the finishing phase, they received a diet containing sorghum silage and concentrate (50:50) for 61 days. The performance indices evaluated were average daily weight gain (ADG), average daily gain during the period (ADP), final body weight. Economic performance indicators were total revenue, total operating expenses per bushel (COT / @), gross margin and net margin. Average daily gain during the growing phase was similar (P> 0.05) among groups with an average of 746 and 456 grams, respectively, for the period's transitional rainy / dry season. The TOE/bushel recreates the grouping of SD (R$ 69.44) was higher than that of groups GN (R$ 66.75) and GH (R$ 64.86). In the finishing period, ADG was similar (P> 0.05) among groups with an average of 1.55kg. The TOE/bushel of the feedlot finishing were R$ 91.48, R$ 114.15 and R$ 122.78 respectively for SD, GN and GH groups. Considering the analysis of the short-cycle system, we obtained the lowest TOE/bushel for grouping SD (R$77.92) followed by grouping GN (R$ 74.89) and GH (R$ 78.43). These results provided higher profitability for the crosses SD (21.99%) compared to other intersections (18.92% and 18.31% respectively, and for GN and GH). In the conditions evaluated the SD group was the most economically efficient.

ABSTRACT

The aim of this study was to determine the yield and the chemical composition of essential oils from Cymbopogon citratus (DC.) Stapf. (lemongrass) and Cymbopogon nardus (L.) Rendle (citronella), and to evaluate their antilisterial activity when pure and combined. The essential oils were obtained from fresh leaves through hydrodistillation by using a modified Clevenger-type device. Yield was calculated based on the extracted mass and the plant humidity. The qualitative analysis was done by gas chromatography-mass spectrometry and the levels of the chemical components were detected by gas chromatography with flame ionization detector. The bacteriostatic effect of different concentrations of pure and combined essential oils was determined by the agar diffusion technique. Minimum inhibitory concentrations (MICs) were used to determine the time of bactericidal action. The yield was 1.39 percent (v w-1 dry basis) for C. citratus essential oil and 2.27 percent (v w-1) for C. nardus essential oil. Geranial and neral were the major components of C. citratus essential oil, while citronellal, geraniol and citronellol were predominant in C. nardus oil. Most of the used concentrations were efficient against Listeria monocytogenes and bactericidal effects were observed for the MICs. Essential oils of C. citratus and C. nardus, pure and combined, constitute new alternatives of natural antibacterials for the control of L. monocytogenes in the food industry.

ABSTRACT

Objective: To investigate the effect of shoulder exercises during radiotherapy in relation to prevention of locoregional physical complications: limitation of range of motion (ROM) and functional capacity, arm circumference and scar tissue adhesion. Methods: Sixty-six women without shoulder ROM impairment following breast cancer surgery that included complete axillary dissection were allocated to one of two groups: 32 in the physical therapy group (PG) (52.7±10.2 years), who underwent an exercise program; and 34 in the control group (CG) (48±10.1years). Shoulder ROM, upper-limb circumference and functional capacity and scar tissue adhesion were evaluated at the beginning and end of radiotherapy and six months after completing radiotherapy. Results: The PG showed improvements in flexion and abduction ROM between the first and third evaluations (flexion from 164.77°±8.9° to 167.98°±9.5° and abduction from 168.56°±10.0° to 175.62°±10.2°),which was not observed in the CG (flexion from 167.06±06° to 165.16°±9.2° and abduction from 169.71° 10.1° to 169.53°±12.8°). There was a statistically significant increase in ROM in the PG in relation to the CG (flexion, p=0.02; and abduction, p=0.004). The circumference and functional capacity were similar between the groups and the frequency of scar tissue adhesion in the CG was twice that observed in the PG (48% versus 24%, p=0.04). Conclusions: These results suggest that shoulder exercises favor maintenance of flexion and abduction ROM of the shoulder and minimize the incidence of scar tissue adhesion in women undergoing radiotherapy for breast cancer treatment.

ABSTRACT

The prevalence of small ruminant lentivirus (SRL) infection was evaluated in goats and sheep in two counties of Pernambuco State, Brasil. Seriological examinations were performed from a total of 672 goats and 325 sheep, one year of age and older, at two abattoirs. Sera were analyzed by agar gel immunodiffusion using Maedi-Visna K-1514 antigens. There were 42 reactive samples (95 percent confidence interval 3.6 percent to 4.9 percent) in both slaughter houses. In São Lourenço da Mata county, 3.2 percent and 4.0 percent of goat and sheep sera were responsive, whereas in Paulista county, 5.1 percent of goat samples and 8.2 percent of sheep were reactive. Thus, the prevalence of small ruminant lentivirus was low in goats and sheep sampled from the region.

ABSTRACT

We bacteriologically analyzed 156 species of Enterobacteriaceae, isolated from 138 patients with community-acquired diabetic foot ulcers, in a prospective study made at a diabetic center and at the Federal University of Ceará, Brazil, from March, 2000, to November, 2001.The samples were cultured using selective media, and identification, susceptibility tests and detection of plasmid-mediated-extended-spectrum-beta-lactamase (ESBL) producing strains were made with conventional and automated methods. The most frequently occurring pathogens were K. pneumoniae (21.2 percent), Morganella morganii (19.9 percent) and E. coli (15.4 percent). High resistance rates were noted for ampicillin, first generation cephalosporin, trimethoprim/sulfamethoxazole, tetracycline, amoxicillin-clavulanic acid and chloramphenicol. ESBL-producing strains were detected in 6 percent of the patients. Resistance among gram-negative bacteria has become increasingly common, even in community-acquired infections

ABSTRACT

The toxic effects of cadmium (2 mug/ml) on membrane lipids and growth of Euglena gracilis were studied using autotrophic (AUTO), heterotrophic (DARK) and mixotrophic (LIGHT) cells. Cadmium caused inhibition of cellular proliferation (IC50 1.2 mug/ml) and morphological alterations which were most pronounced in chloroplasts. The chlorophyll content of LIGHT cadmium-treated cells was reduced 42.5 per cent. Cadmium also caused an increase in protein and total lipid content per cell in all three cell types. Among the membrane lipids, cholesterol content was lower in cadmium-treated cells cultivated under illumination (AUTO: 0.40 ñ 0.02 vs 0.64 ñ 0.08 and LIGHT: 0.40 ñ 0.09 vs 0.53 ñ 0.01 mug/l0(5) cells). There were no changes in total phospholipid content, although cardiolipin content was altered in all three cell types, and in mixotrophic cells there was an increase in phosphatidylglycerol, a phospholipid typically found in chloroplasts. These results suggest that cadmium has an overall toxic effect on Euglena gracilis and that part of the effect can be ascribed to defects in the structure of chloroplasts and mitochondrial membranes.

ABSTRACT

1. The correlation between neurotransmitter substances in specific cell types has renewed interest in the morphology of amacrine cells. In this paper we describe the morphology of two types of amacrine cells in Golgi-stained chick retinas. 2. The first cell type was classified as an asymmetric bistratified amacrine cell suggested to play a role in the formation of complex ganglion cell receptive fields. 3. The second was classified as a bistratified amacrine cell. Their processes were stratified at sublayers 1 and 4 of the inner plexiform layer and their morphological features were similar to those of dopaminergic cells in the chick retina.

ABSTRACT

We have determined the phospholipid composition of E. gracillis under different environmental conditions. Half of the phosphoglycerides was phosphatidylcholine, regardless of growth conditions, but differences were noted in the distribution of the remaining lipids. Autotrophic, green, light-grown euglenas had 31% more amino-containing lipids (phosphatidylethanolamine and phosphatidylserine) than the heterotrophic, white, dark-grown cells. In contrast, dark-grow cells had 47% more anionic lipids, such as the ones belonging to the "phosphatidynositol cycle" (phosphatidylinositol, phosphatidylinositol-phosphate phosphatidylinositol-bis-phosphate and phosphatidic acid) plus cardiolipin. ABSTRACT
